Project description:
Lipid nanoparticles (LNPs) have emerged as a pivotal delivery system for mRNA-based therapeutics. A critical quality attribute of a LNP formulation is the encapsulation efficiency i.e., how much of the formulated mRNA that is associated with the LNP, since it directly impacts safety and efficacy. High encapsulation ensures stability and maximizes cellular uptake of mRNA. However, the encapsulation state of mRNA, describing where the RNA is located and how accessible it is, remains a challenge to elucidate, and the optimal analytical approach to accurately characterize this state has yet to be determined. This project aims to elucidate the mechanisms of mRNA encapsulation in LNPs and the factors during formulation that affects it. To understand the mechanism, the student will  formulate and characterize LNPs using both established analytical methodologies such as: Ion exchange chromatography (IEX) and fluorescent assays (RiboGreen). The graduate will also explore alternative methods, including capillary gel electrophoresis (CGE) and qPCR,  to assess their suitability for analyzing the encapsulation state of mRNA in LNPs. Statistical methods will be used to evaluate the results.

The project will include a systematic comparison of analytical methods to develop a deeper understanding of mRNA encapsulation in LNPs, which could enhance the design and efficacy of future mRNA therapeutics.

Essential Requirements:

  • Ongoing master studies in analytical chemistry, organic chemistry, pharmaceutics, bioprocess technology, process chemistry or biochemistry.
  • Interest in analytical science and technically skilled to perform work in the lab.
